When her parents divorced at an early age, the mother took custody of Sherri and her three sisters and moved with them to California. An experience watching a comedian do a standup act inspired Shepherd to step behind the microphone. As an actress, she debuted on the sitcom Cleghorne!, then landed guest appearances on the aforementioned series. Her father Lawrence was a restaurant manager and her mother LaVerne cleaned homes for a living. She was also raised as a Jehovah's witness. She attended Winston Churchill Elementary School and Eisenhower Junior High School and Hoffman Estates High School.


Sherri started her stand-up career in 1990 while working in Beverly Hills as a legal secretary. Her first TV role was on the show, Cleghorne! Three years later, she continued her career by having recurring roles in television shows such as Everybody loves Raymond, Living Single, Suddenly Susan and The Jamie Foxx Show. Her most notable television role was Ramona Platt from Less Than Perfect. She co-starred for four seasons. She also has a recurring role as Tracy Morgan's wife in 30 Rock. Sherri has appeared in many movies. Most recently in the Oscar nominated picture "Precious". Sherri also co-starred in Dreamworks’ animated feature Madagascar: Escape 2 Africa, where she voiced the character of Ben Stiller’s mother. She starred opposite Queen Latifah in MGM’s Beauty Shop, and appeared with Bernie Mac in Guess Who, as well as, other films such as Who’s Your Caddy and Cellular. Sherri still performs stand up comedy from time to time. On Monday September 10, 2007, Sherri was announced as a permanent co-host of the Day time talk show, The View. Sherri and her fellow co-hosts won a Day Time Emmy Award for their work on The View. Although The View has been a great stepping stone for Sherri, she has recieved backlash for some comments and opinions made. For example, Sherri was crititized for saying that she didn't believe in evolution. And when asked if the world was flat, Sherri responded by saying "I don't know, I'm just trying to take care of my kid". Sherri released a statement following the incident saying this "We were discussing creationism versus evolution, and Barbara asked if I thought the Earth was round or flat. It was just everything coming at me at once, like I was hearing things underwater, and I said, 'I don't know, I'm just trying to take care of my son!' At the end of the show, Barbara goes, 'Sherri, dear, the Earth is round.' I knew that!" She also garnered ridicule after admitting to never voting because she just "never knew the dates or anything"; she stated, "I've never voted for anything in my life." Sherri did however vote for Barack Obama in the presidential election.

Sherri and her husband divorced in 2006 when she discovered he was having an affair. Sherri has said this about her son's disabilty "My son's have developmental delays, but everything that the doctors predicted that he would have, I'm praising God he does not have. He does not have mental retardation; he doesn't have cerebral palsy, no shunts in his brain. But it is a challenge dealing with a child that has developmental delays. You have to have a lot more patience. You know the way our parents raised us, 'Just beat 'em. Give 'em a whuppin!' That doesn't work. You have to find other ways to do things... There are times when I sit on the couch and cry. I go, 'Lord, this is really hard.' But yet, I thank God for really strong girlfriends in my life, who raise children, who say, 'You can do this, and you are a good mother.'" Sherri's autobiography “Permission Slips: Every Woman’s Guide to Giving Herself a Break,” was launched by Grand Central Publishing in October 2009. She attributes her success to her faith in God, her grandmother, and her father’s constant encouragement to become anything she wished through hard work and humor. All of her free time is spent with her son.
