He spent most of the '80s performing in clubs, theaters, and college campuses. He joined the cast of Lorne Michael's long-running television show Saturday Night Live in 1990 as a writer and a performer. On the show, he soon gained popularity for such recurring sketches such as "The Hollywood Minute" in which Spade would sarcastically shred some of Tinseltown's biggest stars with his nasty comments. Spade also proved an able impersonator of celebrities ranging from Jeff Foxworthy to Tom Petty. Spade has appeared on many television talk shows and guest starred on several series. He began his film career in the late '80s playing a small role in Police Academy 4: Citizens on Patrol. In the '90s, he began playing major supporting roles in such films as Coneheads (1993) and P.C.U. (1994). He and former SNL alumni Chris Farley shared top billing in two popular comedies, Tommy Boy (1995) and Black Sheep (1996). Born in Birmingham, Michigan, Spade is the youngest of Wayne Spade and Judy Todd's three sons.

His father moved the family to Scottsdale, Arizona, but abandoned them not long afterwards. His brothers are Bryan Spade and Andy Spade, husband of famed designer Kate Spade and CEO of Kate Spade New York.

Spade's mother eventually remarried, but David's stepfather committed suicide in 1981 (David was 15 years old at the time).

David Spade graduated one year early (he was 17) from Saguaro High School in 1982. He then went on to Arizona State University, where he earned a degree in business in 1986. He was a member of the Sigma Alpha Epsilon fraternity. ASU has a long-running improvisational comedy group that performs for free in the Memorial Union during Friday lunchtimes called "Farce Side Comedy Hour" (from 1985 through 2005, along with a break-off group that peforms on Thursdays).

David Spade is said to have been an early leader in the group recognized for his star potential.

More recently he was the star of the Comedy Central show "The Showbiz Show with David Spade" (2005).In 2005 wherein he more or less resurrected his obnoxious, razor-tongued gossipmonger from the old "Hollywood Minute" put-down segment on SNL, as well as co-starring in the adult-oriented ensemble sitcom "Rules of Engagement" (2007).

On August 26, 2008, David Spade became a father when his ex-girlfriend Jillian Grace gave birth to a baby girl.
